Review: Onto better TRAILs for cancer treatment
Tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL), a member of the TNF cytokine superfamily, has been described to be capable of inducing apoptosis selectively in cancer cells. This review summarizes second-generation novel formulations of TRAIL and other TRAIL-R agonists, which exhibit enhanced cytotoxic capacity toward cancer cells. PEGylated versions using site-specific N-terminal PEGylation of izTRAIL showed widely improved pharmacokinetic profiles in vivo and, furthermore, greatly augmented stability and solubility under physiological conditions.
